The upgrade to WordPress 2.7 Release Candidate 1 took all of five minutes and was (mostly) painless. We use the WordPress plugin for memcached which caused some funky behavior during the upgrade process, but this was easily fixed by temporarily disabling the cache, performing the upgrade and then once more enabling the cache. The new dashboard interface is fantastic: Much better than the 2.6 dashboard. Very clean and usable.
But as with all upgrades, especially to a release candidate, there may be some issues. Please let us know if you find anything… broken.
